I love the theatre, and use the Mayflower often.
Before I start, it is an old building so expectations should bear that in mind.
That said, I find it quite OK for access (I use an electric wheelchair). You can now-a-days get into the ticket hall, and there is level access to almost all of the theatre – main stalls, the circles. There are wheelchair spaces at most of the levels (albeit they are all at the back of the seating areas so can be frustrating when people stand up!). But the spaces are big enough and are raised a little to help viewing the stage.
There is a new area with good wheelchair accessible toilets and automatic doors.
The restaurant is very good also (and expensive!) Accessible via a lift – I’ve only been there once, but I remember it was fine for access.

So providing you don’t mind sitting at the back if you are a wheelchair user – you will find it a pleasant experience.
